The size of this diffraction pattern and the crucial volume it defines is controlled by the numerical aperture of the system’s objective lens and the wavelength of the laser used. Here, the size of the scanning volume is single-minded by the stain size of the optical logic since the image of the scanning laser is not an infinitely small top but a three-dimensional diffraction pattern. Inside CLSM a specimen is illuminated by a top metrologic barcode scanner laser source, and all volume element is associated with a discrete scattering or fluorescence intensity. It varies with the logic optical design, but working distances from hundreds of microns to several millimeters are predictable. The distance from the objective lens to the go up is typically comparable to with the intention of of a square optical microscope.
